Er begann seine Karriere bei verschiedenen Theatern und tourte durch die USA und gar durch Europa.

Bereits 1910 agierte er in ersten Filmen wie "The Fugitive" (10), "Sunshine Sue" (10) und "A Child's Stratagem" (10), doch seine eigentliche Filmkarriere begann mit der Vertragsunterzeichnung bei Biograph 1913.

In den kommenden Jahren verkörperte er unzählige Hauptrollen und er avancierte in kürzester Zeit zu einem begehrten Star beim Publikum.
Zu seinen bekanntesten Filmen dieser Jahre gehören "Strongheart" (14), "The Broken Rose" (14), "His Poor Little Girl" (15), "The Little Runaways" (15), "Harvest" (15), "The Chain of Evidence" (16), "The Iron Will" (16), "The Eyes of Love" (16), "Her Primitive Man" (17), "The Flame of Youth" (17), "Madame Spy" (18) und "The Merry-Go-Round" (19).

In den 20er Jahren blieb seine Popularität ungebrochen und er verkörperte zahlreiche Rollen. Dazu gehören "Miss Hobbs" (20), "Sleepwalker" (22), "Flesh and Blood" (22), "Pirates of the Deep" (22), "The Law of the Sea" (22), "The Wolves of the Waterfront" (23), "The Bad Man" (23), "Into the Net" (24), "The Naked Truth" (24), "She Wolves" (25), "The Far City" (26), "See You in Jail" (27), "Waterfront" (28) und "Children of the Ritz" (29).

Jack Mulhall wurde in dieser Zeit ein reicher Mann, doch der Börsencrash 1929 vernichtete sein Vermögen nahezu vollständig.

Mit über 40 Jahren gehörte er nicht mehr zu den jungen Helden und obwohl er in den vergangenen Jahrzehnten zu den grössten Stars zählte, scheute er nicht, in den kommenden Jahren nebst grossen Rollen auch kleinere Rollen zu verkörpern, um seinen Lebensunterhalt weiterhin verdienen zu können.
Zu seinen bekanntesten Filmen der 30er Jahre gehören "Reaching for the Moon" (30), "Passport to Paradise" (32), "The Three Musketeers" (33), "The Mystery Squadron" (33), "Cleopatra" (34), "Evelyn Prentice" (34), "The Woman in Red" (35), "The Fighting Lady" (35), "The Informer" (35), "The Glass Key" (35), "His Night Out" (35), "Strike Me Pink" (36), "Show Boat" (36), "The Last Outlaw" (36), "Charlie Chan at the Race Track" (36), "Topper" (37), "The Spy Ring" (38), "Of Human Hearts" (38), "Flash Gordon's Trip to Mars" (38), "The Gladiator" (38), "Buck Rogers" (39) und "It's a Wonderful World" (39).

Langsam reduzierte Jack Mulhall seine filmischen Arbeiten, was sich in erster Linie in der Grösse der Rollen denn in der Anzahl der Filme niederschlug.
In den 40er Jahren agierte er erneut in zahlreichen Filmen, ebenso in den 50er Jahren, wo in seinem letzten filmischen Jahrzehnt vor allem das Fernsehen den Grossteil seiner Arbeiten ausmachte.

Zu seinen bekanntesten Filmen dieser Zeit gehören "The Son of Monte Cristo" (40), "Dangerous Lady" (41), "It Started with Eve" (41), "A Gentleman After Dark" (42), "Foreign Agent" (42), "The Glass Key" (42), "The Ape Man" (43), "Lady in the Dark" (44), "Dillinger" (45), "Monsieur Beaucaire" (46), "Lulu Belle" (48), "Chained for Life" (51), "The Man with the Golden Arm" (55), "Around the World in Eighty Days" (56) und "The Atomic Submarine" (59).

Nach seinem Rückzug als Schauspieler vermittelte er für die "Screen Actors Guild" Schauspieler, eine Tätigkeit, die er bis 1974 ausübte.
